چکیده
Background: Metabolic pathway operation and NAPDH homeostasis in non-growing bacteria is unknown. Results: Newly discovered metabolic cycles and known metabolic reactions balance concertedly the catabolic NADPH production. Conclusion: We propose the first quantitative NADPH balancing model under non-growing conditions. Significance: NADPH balancing is significantly different between resting and growing bacteria, reflecting microbial survival strategies during environmental challenges.

Different biochemical mechanisms ensure network-wide balancing of reducing equivalents in microbial metabolism.
To sustain growth, the catabolic formation of the redox equivalent NADPH must be balanced with the anabolic demand. The mechanisms that ensure such network-wide balancing, however, are presently not understood. Based on 13C-detected intracellular fluxes, metabolite concentrations, and cofactor specificities for all relevant central metabolic enzymes, we have quantified catabolic NADPH productio...
متن کاملMetabolic fluxes during strong carbon catabolite repression by malate in Bacillus subtilis.
Commonly glucose is considered to be the only preferred substrate in Bacillus subtilis whose presence represses utilization of other alternative substrates. Because recent data indicate that malate might be an exception, we quantify here the carbon source utilization hierarchy. Based on physiology and transcriptional data during co-utilization experiments with eight carbon substrates, we demons...

The energetic efficiency of microbial growth is significantly reduced in cultures growing under glucose excess compared to cultures growing under glucose limitation, but the magnitude to which different energy-dissipating processes contribute to the reduced efficiency is currently not well understood. We introduce here a new concept for balancing the total cellular energy flux that is based on ...
متن کاملRole of the C-terminal extension stacked on the re-face of the isoalloxazine ring moiety of the flavin adenine dinucleotide prosthetic group in ferredoxin-NADP(+) oxidoreductase from Bacillus subtilis.
Ferredoxin-NADP(+) oxidoreductase [EC 1.18.1.2] from Bacillus subtilis (BsFNR) is homologous to the bacterial NADPH-thioredoxin reductase, but possesses a unique C-terminal extension that covers the re-face of the isoalloxazine ring moiety of the flavin adenine dinucleotide (FAD) prosthetic group. In this report, we utilize BsFNR mutants depleted of their C-terminal residues to examine the impo...
متن کاملFlux Distribution in Bacillus subtilis: Inspection on Plurality of Optimal Solutions
Linear programming problems with alternate solutions are challenging due to the choice of multiple strategiesresulting in the same optimal value of the objective function. However, searching for these solutions is atedious task, especially when using mixed integer linear programming (MILP), as previously applied tometabolic models. Therefore, judgment on plurality of optimal m...
